Individual determination of ploidy level in experimentally induced triploid bivalve molluscs using image cytometry. The sexual maturation in bivalve molluscs causes decreased growth and sometimes increased mortalities. These drawbacks can be avoided by genetic sterilization (triploidy). Triploïdy induction require a egg treatment and a ploidy control at each generation. The ploidy level is usually determined by counting the chromosomes, a technique which is relatively easy for the trochophora stage and for juveniles measuring from 5 mm to 3 cm only. ln order to be able to follow up the evolution of the ploidy rate during any stage of animallife, we have developed an estimation method based on image cytometry. Essais d'obtention de lignées pures de mollusques par gynogenèse ArchiMer

La gynogenèse est le développement d'un oeuf activé par un spermatozoïde, mais sans participation du génome paternel. Sur le plan génétique, elle est équivelente à la parthénogénèse mais elle est déclenchée par d'autres facteurs. Chez l'huître creuse, Crassostrea gigas, l'inactiviation génétique du sperme par les UV est plus difficile à obtenir que chez les poissons, principalement du fait d'une grande variabilité inter-indivuelle de la motilité avant irradiation. Par ailleurs, l'absence constatée d'effet Hetwig rend plus délicate la définition d'un temps optimal d'exposition aux UV. Les haploïdes obtenus sont anormaux et n'atteignent pas le stade de larve D. La diploïdie peut être efficacement restaurée en retenant un globule polaire grâce au traitement...

Following a phase of experimental system development, acquisition and improvement of triploidisation techniques carried out in 1989, the 1990 program divides into four main actions:1) significant production of diploid and triploid populations of cupped oysters and Pacific clams in order to compare their performances in French coastal waters2) first trials of triploidy induction in European clam3) development of a ploidy control technique at any stage of the animal life, through digital imaging4) summary of the first trials of tetraploidy induction in C. gigas and R. philippinarum

The third part of the polyploidisation program of the main French species of commercial interest, this report presents the actions and results for the year 1991 which was devoted to:- continuation of the development trials of the triploidy induction technique in Ruditapes decussates (i.e. indigenous clam)- production of diploid and triploid populations of Ostrea edulis (i.e. flat oyster)- control of diploid and triploid population performances in cupped oyster and Pacific clam in their natural habitat- continuation of the development of a ploidy control technique through image analysis- beginning of the development of a new polyploidy induction technique in bivalve molluscs
